After 14 days of administration of OSI-906, the OSI-906 group showed hyperglycemia, body weight loss, and decreased weight of quadriceps and gastrocnemius muscles. Tissue analysis showed a significant increase in the number and proportion of oxidative fibers in the tibialis anterior muscle after OSI-906 administration, indicating that inhibition of IR and IGF1R induces plastic changes in fully differentiated skeletal muscle. Genetic analysis indicated OSI-906 promotes glucose uptake in skeletal muscle, suppress cell cycle control, promotes proteolysis, and enhances autophagy., 18K08484

Distinct β cell subpopulations potentially have unique functional identity. We topologically divided pancreas into a head (right side, nestled in the curve of the duodenum), body, and tail (left side in close proximity with the spleen) region in mice. Then, we compared head islets with tail islets histologically and functionally. Tail islets contained significantly higher proportion of α-cells. Glucose-stimulated insulin secretion was significantly greater in tail islets than in head islets, while β cells in head islets showed higher response to GLP-1. The results of gene expression microarray analysis and proteomics analysis indicated that some functional genes including pancreatic progenitor markers were differentially expressed between head and tail islets. We now systematically investigated changes in the expression of genes involved in beta cell function and proliferation in mouse islets stimulated with glucokinase activator (GKA). GKA-stimulated IRS2 production affected beta cell proliferation but not beta cell function. Specifically, we focused the roles of fibulin 5, EGFR and S100A8 in beta cells. Of note, both glucotoxicity and lipotoxicity triggered S100A8 secretion from pancreatic islets, which in turn promoted macrophage infiltration of the islets. In this study, we investigated the impact of glucokinase activation by glucokinase activator (GKA) on ER stress in beta-cells. GKA administration improved beta-cell apoptosis in Akita mice, a model of ER stress-mediated diabetes. IRS-2-deficient islets were vulnerable, but bIRS-2-Tg islets were resistant to ER stress-induced apoptosis. Meanwhile, GKA regulated the expressions of C/EBP homologous protein (CHOP) and other ER stress-related genes in an IRS-2-independent fashion in islets. GKA suppressed the expressions of CHOP and Bcl2-associated X protein (Bax) and protected against beta-cell apoptosis under ER stress in an ERK1/2-dependent, IRS-2-independent manner.
