  • Understanding the mechanisms of epileptogenesis in the hippocampal dentate gyrus using in vivo calcium imaging., Jinde Seiichiro, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), The University of Tokyo, 01 Apr. 2018, 31 Mar. 2021, Mossy cells in the hippocampal dentate gyrus are known to be deeply involved in the pathophysiology of temporal lobe epilepsy, but the exact role of mossy cells has not been elucidated. In the present study, the activity of mossy cells was visualized during epileptic seizures in the awake state by combining the mice that can be genetically engineered specifically for mossy cells with in vivo calcium imaging. The results showed that the activity of mossy cells was largely affected by increased activity along with behavioral epileptic seizures, and that, when the mossy cells was excited, the epileptic activity of granule cells in the dentate gyrus was increased. These studies have revealed a part of the mechanisms of epileptogenesis in temporal lobe epilepsy, and are further expected to lead to the development of new therapies in the future., 18K07589

Specifically, by using magnetic resonance imaging, we conducted longitudinal measurements of mouse brains (C57BL/6J) and thereby explored differential characteristics of the structure and spontaneous activity between autism model and wild-type mice. Conducting voxel-based morphometry on the structural images, inter-regional functional connectivity analysis on the resting-state functional images, as well as comparison of these findings with independent human studies, we have identified that gray matter volume and functional connectivity involving the limbic system through basal ganglia are candidate biomarkers of autism that are common to both human and mouse. Future investigation on their relationship with autism-specific behavioral phenotypes (such as socio-communicative deficits) is required to further clarify the clinical implication., 25461752

This strain showed the reduced emotional reactivity and the decreased hippocampal neurogenesis. In this study, we examined whether a perinatal exposure to a low dose BPA modulates the stress sensitivity including the functions of HPA-axis and neurotrophic factiors in later life. In female rats, the serum corticosterone (CORT) level of BPA+ rats was significantly higher than BPA-rats under basal conditions. No difference was observed in CORT level immediately after the immobilization stress, whereas CORT level of BPA+ rats was significantly higher than BPA-rats 12 h after the immobilization stress. In male rats, there was no difference between BPA+ and BPA-rats in the serum corticosterone level under basal conditions nor after the immobilization stress. No difference was detected in the expression of GR, BDNF and c-fos mRNA between BPA+ and BPA-rats before treatment nor after a stress. 2.Maternal separation (MS) model : In experimental animals, maternal separation (MS) during neonatal period was found critical for susceptibility to stress in adult offspring. Differential gene expression caused by the MS stress was investigated with DNA microarray analysis using the rat hippocampal samples. RT-PCR was also performed to validate the results. Expression of transthyretin (TTR) was drastically reduced in the MS group. It is known that TTR is specifically expressed in the choroid plexus (CP). When normalized by prostaglandin D synthase, TTR still indicated significant decrease. The two other CP-enriched genes, angiotensin I converting enzyme I and insulin-like growth factor II, were also significantly down-regulated in the MS group. Considering that TTR is a carrier protein of thyroid hormone and retinol and that those factors are indispensable for neuronal development and function, significant reduction of TTR should have critical roles in generation of stress susceptibility., 15591211
