  • Research of the effects of the hydrogen bonds derived from difluoromethyl group on selective organic reactions, Amii Hideki, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Challenging Exploratory Research, Gunma University, 01 Apr. 2016, 31 Mar. 2019, The present research deals with the effects of the hydrogen bonds derived from difluoromethyl group on selective organic reactions. The difluoromethyl group has been proposed to act as a hydrogen bond donor to a carbonyl oxygen atom. For the effective introduction of a difluoromethyl group into organic molecules, we discovered selective synthesis of o-(difluoromethyl)benzoates by halogen-lithium exchange of (o-bromophenyl)difluoroacetates followed by quenching under basic conditions. Employing EtOH as a quenching agent for the reactions of (o-bromophenyl)difluoroacetates with n-butyllithium in THF at -78 ℃ afforded the corresponding o-(difluoromethyl)benzoates. Furthermore, the intramolecular hydrogen bonding between the difluoromethyl group and the ester moieties in o-(difluoromethyl)benzoates was observed by 1H NMR and IR spectra.   • Synthesis of conjugated organic compounds using fluorinated isobenzofurans, Amii Hideki, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Gunma University, 01 Apr. 2016, 31 Mar. 2019, Fluorinated isobenzofurans are considered to be promising building blocks for the synthesis of fluorine-containing carbocyclic and heterocyclic systems. We have developed a novel method for the generation of trifluoromethylated isobenzofurans. The cascade trifluoromethylation/intramolecular acetalization of o-phthalaldehyde proceeded smoothly to give the lactol in high yield. The lactol easily underwent dehydration under acidic conditions to generate trifluoromethylated isobenzofuran, which underwent the intermolecular Diels-Alder reactions with alkenes to give the adducts in good yields. The resultant bicyclo compounds are good precursors of functionalized 1-trifluoromethylnaphthalenes. For instance, [4+2] cycloaddition with naphthpquinone and the subsequent transformations gave trifluoromethylated tetracene. Nucleophilic introduction of fluoroalkyl groups is one of the most powerful methods for the synthesis of organofluorine compounds. alpha-Fluorobenzyl anions were generated by deprotonation of benzylic fluorides upon treatment with lithium tetramethylpiperidide (LTMP). Nucleophilic reactions using fluorobenzyl anions with electrophiles led to the formation of monofluoroalkyl and difluoroalkyl compounds. Previously, we reported the catalytic procedure for aromatic trifluoromethylation by the use of trifluoroacetaldehyde hemiaminal derivative as a cross-coupling partner in the presence of 10 mol% of CuI and 1,10-phenanthroline (phen). Afrer undertaking a survey of Cu catalysts, we found that for the cross-coupling reactions with aryl iodides, the use of copper thiophene-2-carboxylate (CuTC) as a source of copper(I) affords much improved results., 26550074

Difluoromethylene compounds have been considerable synthetic targets because of their wide utility. Among them, difluoromethylated aromatic compounds (Ar-CF2-R) have received a great deal of attention in the design of bioactive molecules. Previously, we reported the reaction sequence involving cross-coupling and decarboxylation to afford difluoromethyl aromatics (Ar-CF2H). Herein we developed versatile decarboxylative transformations for the synthesis of aromatic compounds bearing functionalized difluoromethyl groups. Upon heating in DMSO or NMP at 170 °C with CuI, potassium 2-aryl-2,2-difluoroacetates underwent decarboxylation to generate aryldifluoromethyl anions (ArCF2-), which reacted with electrophiles to afford a variety of gem-difluoromethylene compounds in moderate to good yields., 25288044

Meanwhile, a new technology for highly selective synthesis of enantiomerically pure organofluorine compounds has been required. Toward stereoselective C-F bond activation, we examined Lewis acid-mediated defluorinative alkylation using gem-difluoromethylene compounds. Organoaluminum compounds were found to promote C-F bond activation of β-fluoro ethanol derivativess. When β, β-difluoro ethanols were treated with a stoichiometric amount of trimethylaluminum, defluorination and the subsequent methylation proceeded smoothly to give the ketones in moderate yields. For the preparation of several gem-difluoromethylene substrates, we developed the cross-coupling routes to 2-aryl-2,2-difluoroacetates. This review summarizes some approaches to realize complete reversal of enantioselectivity by changing various conditions. Our new approach to achieve reversal of enantioselection in the reaction of enantioselective addition of diketene to aldehydes affording optically active 5-hydroxy-3-ketoesters in high optical yield using Schiff bases containing oxazoline moiety derived from L-serine, was achieved. Starting from one chiral center (in this case from L-serine), we obtained two chiral Schiff base possessing oxazoline moieties, each of which recognized different enantioface of aldehydes in the addition reaction of diketene to aldehydes in high level (up to 93% ee R and 89% ee (S), respectively).   • Creation of Fluorinated Amino Acids Library, UNEYAMA Kenji; KATAGIRI Toshimasa; AMII Hideki, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), OKAYAMA UNIVERSITY, 2001, 2003, In order to create a library of fluorinated amino acids, a basic study on magnesium-promoted C-F bond activation has been done and then several fluoorinated amino acids have been synthesized stereoselectively. The followings are a summary of the syntheses. 1) Syntheses of optically active β,β-difluoroproline(1) and β,β-difluoroglutamates(2 α-imino-β bromo-β,β-difluoropropanate was enantioselecdvely hydrogenated by Pd(OCOCF3)2 in trifluoroethanol to provide the corresponding amine in 87%ee, which was transformed to (1) and (2) by sequensial chemical reactions. 2) Syntheses of optically active α-trifluoromethyl-α-carboalkoxyaziridine (3) Optically active α-trifluoromethylaziridine was lithiated in THF and carboalkoxylated with chloro formate in 95% yield with retention of the configuration. The aziridine could be ring-opened by the nucleuophiles leading to the general syntheses of α-trifluoromethylated amino acids. 3) Synthese of N, α-C-disilyl-β,β-difluoroenamines (4) and their utilization for fluorinated amino acids Mg-promoted C-F bond activation of trifluoroacetimidoyl chloride in THF provided of N, α-C-bissilyl-β,β-difluoroenamines in excellent yields, which are synthetic equivalents of N, α-C-and α-C-β-C-dianion. The bissilyl enamines could be allcylated stepwise leading to a vaiety of functionalized amino acid precursors. 4) Syntheses of optically active trifluoromethylnorcoronamc acids (5) The intramolecular stereospecific substitution of hydroxy group with cyanide-stabilized carbanion provided optically active trifluoromethylcyclopropanes which could be transformed to amino acids (5). 5) Syntheses of trifluoromethylisocysteine(6) Condensation of trifluoroacetimidoyl chloride with S-protected α-thioacetate followed by disstereoselective reduution of imono moiety and deprotection of thio moiety gave (6). 6) Rhodium catalyzed quinoline ring formation by the reaction of trifluoroacetimidoyl chloride with acetylene carboaylates provided quinoline carboxylates in one-pot reaction., 13555254

In order to develop a practical synthetic method for the cyclophane, Mg-promoted C-F bond activtion has been extensively studied. And one of the protocol for the purpose has been found which consists of reaction of bistrifluoromethyl benzene wirh magnesium in the presence of trimethylsilane in DMF. The reaction provided the desired trimethylsilylated benzyl compound in 50-60% yields. Fluroride-catalyzed desilylated defluorination gave the desired cyclophane via quinodimethane in 53% yield. The present method will be competitive to the Dolbier method based on Zn-metal promoted dechlorinative preparation from the point of availability of bistrifluoromethylbenzene., 12450356

In particular, difluoroenolsilyl ethers, difluoroenamines, and 4'-trfluoromethy1, 1-difluorobenzy1 trimethylsilane nave been effectively prepared. The reductive reaction proceeded very smoothy1 by the action of two mole equivalents of metallic magnesium and four mole equivalents of TMS-C1 at 0℃ for 30 min. The industrially feasible production of difluorovinyl monomer for difluorovinyron and octafluoro [2.2 paracyclophane for flurorinated perarylene polymer would be realized. 2) Synthesis of optically active fluorinated amino acids Asymmetric reduction of imino group of 3, 3, 3-trifluoro-2-iminopropanoates was achieved by palladium catalyzed reduction in 3, 3, 3-trifluoroethanol which played an essentially important role for the asymmetric hydrogenation. Syntheses of perfluoroalkylated α-diimines, 2, 2, 3, 3,-tetrafluoro-1, 4-diketones and pinacols. Novel trifluoro- and perfluoroalkyl-α-diimines have been efficiently prepared by the palladium catalyzed coupling reaction of the corresponding imidoyl halides. The diimines could be a promising catalyst for polymerization of alkenes which would be studied in future. Likewise, difluoroenolsilyl ethers and trifluoromethyl ketones have been coupled by Cu (II) oxidation and Zn reduction, respectively., 09305058
