    CAPS proteins are known to associate with secretory vesicles and regulate their secretion. I have shown that CAPS2 is involved in the secretion of BDNF (brain-derived neurotrophic factor), which is important for the formation of neural networks (Sadakata et al., J. Neurosci., 2004; J. Comp. Neurol., 2007; Shinoda, Sadakata et al. Proc. Natl. Acad. Sci. USA., 2011).
    Expression analysis of human CAPS2 revealed that abnormal splicing occurs in some autistic patients (Sadakata et al., J. Clin. Invest., 2007). It was suggested that BDNF secretion is abnormal in brains expressing autistic-type CAPS2 protein, resulting in abnormal neural network formation (Sadakata et al., J. Clin. Invest., 2007; J. Neurosci., 2007). To prove this hypothesis, I generated genetically engineered mice that express only the autistic CAPS2 protein. These mice exhibited various autism-like traits, including abnormal social behavior (Sadakata et al., Proc. Natl. Acad. Sci. USA., 2012). Analysis of CAPS2 heterozygous mice revealed that a decrease in CAPS2 protein also causes autism (Sadakata et al., FEBS Lett., 2013).
    The CAPS2 protein was also found to bind to class II ARF proteins, indicating that CAPS and class II ARF proteins are involved in the formation of secretory vesicles in the Golgi apparatus (Sadakata et al; FEBS J., 2012; J. Neurosci., 2013; Neurosci. lett., 2017). We have also shown that Class II ARF proteins transport specific Na+ channels to neuroaxonal origins and are involved in essential tremor (Hosoi et al., J. Neurosci., 2019).
