  • Epigenetic modulation by chemical substances on thyroid hormone-mediated brain development., Koibuchi Noriyuki, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Gunma University, 01 Apr. 2018, 31 Mar. 2022, The aim of this study was to investigate the effect of environmental chemicals on the thyroid hormone (TH)-mediated signal transduction using various levels of experiment starting from in silico docking analysis to behavioral analysis. The hydroxylated PCB, which was originally to be used, became unavailable due to restriction in Japan. Instead, isoflavones, PFOS, gadolinium contrast agents, etc. were used. In isoflavones, genistein, daidzein and its metabolite equol exerted their action by crosstalk with several receptors such as TH and estrogen receptors and promoted neurite development and astrocyte migration to affect brain development. For PFOS, its exposure caused various abnormal behaviors and suppressed long-term depression of Purkinje cells, which are target cells for TH. Furthermore, the gadolinium contrast agents also acted by crosstalk of multiple receptors.   • Mechanism of activity-dependent motor improvement in SCA, Hosoi Nobutake, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Gunma University, 01 Apr. 2017, 31 Mar. 2022, In this study, I focused on congenital spinocerebellar ataxia (SCA) which is characterized by cerebellar dysfunction, and the purpose of the study was examining the mechanism by which oral administration of low-dose baclofen may improve motor performance of SCA model mice in an activity-dependent manner. To this end, I tried to develop a technique for activity-dependent labeling of cerebellar neurons, but it turned out to be difficult because of the unstableness and the low efficiency of activity-dependent labeling in the cerebellum. The effect of low-dose baclofen on cerebellar function was examined, and the results suggest that baclofen might act on the brain regions other than the cerebellum to improve motor performance of SCA3 model mice., 17K07103
  • Development of gene therapy against neurodegenerative diseases using CRISPR/Cas9-based genome editing, Hirai Hirokazu; HOSOI Nobutake; KONNO Ayumu; MATSUZAKI Yasunori, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Challenging Exploratory Research, Gunma University, 01 Apr. 2016, 31 Mar. 2018, In this research, we aimed to establish a technique that allows elimination of a disease-causing gene from specific cell populations in the brain, using adeno-associated virus (AAV) vectors in combination with CRISPR/Cas9 system. Initially, we characterized neuron-specific, astrocyte-specific and cerebellar Purkinje cell-specific promoters available for AAV vector-mediated transgene expression. The results were published in 3 papers. Then, using the AAV vectors, we examined whether a gene responsible for developmental defects of the brain could be knocked out from Purkinje cells by CRISPR/Cas9 system.   • Elucidation of cellular and nuclear signals that regulate motor learning, Hirai Hirokazu; HOSOI Nobutake; KONNO Ayumu; MATSUZAKI Yasunori, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Gunma University, 01 Apr. 2015, 31 Mar. 2018, Decrease of several genes including transient receptor potential cation channel type 3 (TRPC3), inositol trisphosphate (IP3) receptor and Homer protein homolog 3 (HOMER3) were found in the cerebellum of spinocerebellar ataxia type 1 (SCA1) knock-in mouse. We identified retinoid-related orphan receptor α (RORα), which is a transcription factor that could regulate expression of those genes. Moreover, we found that a gene “X” is increased more than 3 times in SCA1 knock-in mice 3 h after treatment with baclofen and rotarod training. Since haploinsufficiency of the gene “X” is known to cause mental retardation, upregulation of the gene “X” may play a key role in baclofen-induced improvement of motor learning in SCA1 mice., 15H04254

Originally, in order to examine functional roles of cerebellar glial ATP receptors, I tried to knockdown glial ATP receptors using microRNA-mediated gene silencing technique and it did not work. However, while I tried to express transgenes selectively in cerebellar glial cells with adeno-associated virus (AAV) vectors, I managed to develop a method in which larger transgenes can be expressed in glial cells, compared to the usual usage of AAV vectors. To elucidate the functional role of Ca-permeable AMPA receptors in cerebellar glial cells, I attempted to develop a method in which glial Ca-permeable AMPA receptors are turned Ca-impermeable during cerebellar development.
