In this study, we focused on hypoxic microenvironment and regulation of the transcription factor C/EBPbeta, which is activated by BCR-ABL and promotes differentiation and exhaustion of CML-SCs. Hypoxic condition in vitro reduced the expression of C/EBPbeta in CML cell lines, and cytokine treatment could not reverse this effect. In contrast, when the mice, which had been transplanted with BCA-ABL expressing bone marrow cells, were treated with cytokines, exhaustion of CML-SCs were promoted via upregulation of C/EBPbeta. These results suggest that cytokine treatment may eradicate CML-SCs by remodeling bone marrow microenvironment including hypoxia in addition to direct action on CML-SCs., 25461415

The purpose of this study was to examine whether inactivation of PHD2 in the liver played an important role for activating Cori cycle and decreasing lactate levels in circulation. Blood lactate levels after treadmill test or lactate tolerance test were significantly lower in PHD2-liver-specific knockout (PHD2-LKO) mice. The stable isotope labeled lactate incorporation assay revealed that livers of PHD2-LKO mice produced significantly greater amounts of glucose derived from the labeled lactate than controls, suggesting that the blockade of PHD2 in the liver helps improve lactic acidosis by activating gluconeogenesis from lactate., Competitive research funding, 24659154

Hypoxic response is mainly regulated by transcriptional factor HIF (hypoxia-inducible factor), which is also negatively regulated by “oxygen sensor” prolyl-hydroxylase PHDs. Among all PHDs, inactivation of PHD2 alone is sufficient to activate HIF pathway. Given that hypoxic response is designed for tissue protection under the hypoxic environment such as ischemic diseases, activation of HIF by PHD2-blocade should have beneficial role in ischemia/reperfusion model. Here we reported that inactivation of Phd2 in mice myocardial infarction model revealed smaller infarction size and better cardiac function by maintaining higher ATP with lower oxygen consumption. Next, we succeeded to generate NMR-iPS cells and revealed that NMR-iPS cells have tumor-resistance. Interestingly, NMR-fibroblasts failed to reprogram in a hypoxic condition. Thus, the response of the NMR cells to oxygen condition during the reprogramming process seems to be unique. Although their Km values for oxygen differ, the activity of these oxygenases decreases by hypoxia. In this study, we focused on hydroxylase, whose activity has been well studied by hypoxia, and aimed to quantify, evaluate, and understand the hypoxic response itself by quantifying the hydroxylation state in cells, i.e., "hydroxyome analysis".
