CRISPR/Cas9ゲノム編集を用いた神経変性疾患の根本的遺伝子治療法の開発, Development of gene therapy against neurodegenerative diseases using CRISPR/Cas9-based genome editing, 平井 宏和; 細井 延武; 今野 歩; 松崎 泰教, Hirai Hirokazu; HOSOI Nobutake; KONNO Ayumu; MATSUZAKI Yasunori, 日本学術振興会, Japan Society for the Promotion of Science, 科学研究費助成事業, Grants-in-Aid for Scientific Research, 挑戦的萌芽研究, Grant-in-Aid for Challenging Exploratory Research, 群馬大学, Gunma University, 2016年04月01日, 2018年03月31日, CRISPR/Cas9システムと細胞種特異的アデノ随伴ウイルス(AAV)ベクター発現系を組み合わせ、生体脳内において特定の細胞群から疾患遺伝子を取り除く技術を確立することを目的とした。まずAAVベクターに搭載可能なニューロン特異的プロモーターとアストロサイトプロモーター、小脳プルキンエ細胞特異的L7-6プロモーターを開発して論文発表した。さらにプルキンエ細胞特異的発現AAVベクターを用いて、脳の発達異常原因遺伝子を、CRISPR/Cas9ゲノム編集技術によってノックアウトできるか検討した。その結果、小脳全域の約3分の2のプルキンエ細胞において、原因遺伝子をノックアウトすることに成功した。, In this research, we aimed to establish a technique that allows elimination of a disease-causing gene from specific cell populations in the brain, using adeno-associated virus (AAV) vectors in combination with CRISPR/Cas9 system. Initially, we characterized neuron-specific, astrocyte-specific and cerebellar Purkinje cell-specific promoters available for AAV vector-mediated transgene expression. The results were published in 3 papers. Then, using the AAV vectors, we examined whether a gene responsible for developmental defects of the brain could be knocked out from Purkinje cells by CRISPR/Cas9 system. The results showed successful removal of the gene from around two thirds of Purkinje cells., 16K15477
運動記憶を形成する細胞内・核内シグナルの解明, Elucidation of cellular and nuclear signals that regulate motor learning, 平井 宏和; 細井 延武; 今野 歩; 松崎 泰教, Hirai Hirokazu; HOSOI Nobutake; KONNO Ayumu; MATSUZAKI Yasunori, 日本学術振興会, Japan Society for the Promotion of Science, 科学研究費助成事業, Grants-in-Aid for Scientific Research, 基盤研究(B), Grant-in-Aid for Scientific Research (B), 群馬大学, Gunma University, 2015年04月01日, 2018年03月31日, 脊髄小脳失調症1型(SCA1)モデルマウス小脳において発現低下している分子として、TRPC3、IP3受容体、Homer3が見つかって来た。その上流制御分子としてretinoid-related orphan receptor α(RORα)を明らかにした。またSCA1マウスにバクロフェンを投与、続いて運動を行わせ、3時間後に小脳組織をマイクロアレイで解析したところ、エピゲノム修飾に関わる遺伝子Xが3倍以上増加していることがわかった。遺伝子Xのhaploinsufficiencyは精神発達遅滞を引き起こすことが知られている。遺伝子X発現上昇は小脳運動学習能に関連している可能性が考えられた。, Decrease of several genes including transient receptor potential cation channel type 3 (TRPC3), inositol trisphosphate (IP3) receptor and Homer protein homolog 3 (HOMER3) were found in the cerebellum of spinocerebellar ataxia type 1 (SCA1) knock-in mouse. We identified retinoid-related orphan receptor α (RORα), which is a transcription factor that could regulate expression of those genes. Moreover, we found that a gene “X” is increased more than 3 times in SCA1 knock-in mice 3 h after treatment with baclofen and rotarod training. Since haploinsufficiency of the gene “X” is known to cause mental retardation, upregulation of the gene “X” may play a key role in baclofen-induced improvement of motor learning in SCA1 mice., 15H04254
